## Abstract

The original study tackled the highmortality burden of clear cell renal cell carcinoma, focusing on the loss of VHL and consequent HIF2 activation that drives tumour growth and systemic effects such as hypercalcaemia and cachexia, with parathyroidrelated protein identified as a key mediator. Motivated by a desire to clarify the molecular circuitry underlying these clinical manifestations, we reanalyzed RNAseq data from GSE289579. The workflow drew expression counts and sample metadata from ARCHS4, assembled them into an Ann- Data object, applied a differential expression contrast between treated and control cells using limmavoom, and submitted the resulting gene signatures to Enrichr and PerturbSeqr for pathway and perturbagen en- richment. The reanalysis revealed that genes upregulated by the treatment cluster in immunerelated and extracellularmatrix remodeling pathways, with enriched TNF and TGF signalling and a STAT3centric tran- scriptional network, while genes downregulated display a striking depletion of cellcycle, DNAreplication and kinetochoreassembly machinery. PerturbSeqr highlighted a pronounced sensitivity to CDK4/6 inhibition, with palbociclib repeatedly emerging as a mimicker, suggesting that the drugs effects converge on halting proliferative signalling and rewiring the tumour transcriptome toward an inflammatory, matrixremodelling phenotype. This integrated view links HIF2 antagonism to coordinated suppression of growth programs and activation of immune pathways, offering mechanistic insight and potential combination strategies.
