Abstract

Original study examined YAP overexpression in H727 lung carcinoid cells to understand how YAPTEAD signaling influences neuroendocrine differentiation and proliferation. We reanalyzed the same GEO dataset to refine the transcriptional signature and identify potential therapeutic targets. The workflow retrieved the expression counts and sample annotations from ARCHS4, formatted the data into an AnnData object, applied a general differential expression routine to generate a gene set, and submitted that set to an enrichment platform and a perturbation matching service. The reanalysis confirmed that YAP overexpression drives a coordinated upregulation of extracellularmatrix remodeling, focaladhesion, and tightjunction genes, with strong enrich- ment for ERBB2/EGFR signaling and epithelialtube branching pathways. Downregulated genes clustered in developmental and metabolic categories, including heartdevelopmental and centralcarbonmetabolism pathways, suggesting a shift away from differentiation toward a proliferative state. The perturbation matching identified kinase inhibitors such as sorafenib, PD0325901, and dasatinib, as well as the NADmetabolism modulator nicotinamide, as compounds that mimic or reverse the YAPdriven signature, highlighting potential therapeutic angles. This refined view links YAP activity to specific signaling modules and offers actionable insights for targeting lung carcinoid tumors.
